The effects of L-theanine and its 2:1 ratio with caffeine appear in dozens of published studies. Here are ten.

Reviewing multiple randomised controlled trials, the authors concluded that L-theanine and caffeine have synergistic effects on attention and task-switching, with the combination consistently outperforming either ingredient in isolation.
In a triple-blind, placebo-controlled study, participants given L-theanine alongside caffeine showed lower cortisol responses and reduced subjective stress during a standardised psychological stress test, while maintaining the cognitive improvements associated with caffeine.
Participants given 100mg L-theanine and 50mg caffeine showed faster response times and improved accuracy on attention-switching tasks compared to placebo.
A randomised, placebo-controlled trial found 200mg daily L-theanine over 4 weeks was associated with reductions in stress-related symptoms including sleep quality and cognitive function.
Using functional MRI, researchers observed reduced activity in default mode network regions during sustained attention tasks, suggesting reduced mind wandering.
Participants deprived of sleep and given L-theanine with caffeine showed faster reaction times without an increase in errors, compared to caffeine alone.
A 12-week trial in adults aged 50-69 found daily consumption of caffeine and L-theanine was associated with improvements in attention and executive function.
In a double-blind placebo-controlled trial, 200mg L-theanine significantly reduced heart rate and salivary immunoglobulin A responses to an acute stress task, indicating both subjective and physiological stress reduction.
In a double-blind, placebo-controlled crossover study of 34 healthy adults, an L-theanine drink significantly reduced subjective stress one hour post-dose and salivary cortisol three hours post-dose following a cognitive stressor.
A double-blind, randomised, placebo-controlled trial found that 450-900mg daily L-theanine supplementation over 8 weeks was well-tolerated and associated with improvements in anxiety, sleep satisfaction, and cognitive function in adults with generalised anxiety disorder.
